Cognition Therapeutics President & CEO Lisa Ricciardi joins Natalie Stoberman from the Proactive newsroom to discuss the company's unique focus on the treatment of Lewy body Dementia as well as other cognitive diseases like Alzheimer's Disease. Cognition Therapeutics is a clinical-stage biopharmaceutical company engaged in the discovery and development of innovative, small molecule therapeutics targeting age-related degenerative disorders of the central nervous system and retina. The company is currently investigating its lead candidate CT1812 in clinical programs in Alzheimer’s disease, dementia with Lewy bodies (DLB) and dry age-related macular degeneration (dry AMD). #proactiveinvestors #cognitiontherapeutics #NASDAQ #CGTX #biopharma #alzheimersdisease #dementia #DLB #lewybodydementia #invest #investing #investment #investor #stockmarket #stocks #stock #stockmarketnews
CT1812 is a small molecule sigma-2 receptor (S2R) modulator in development for Alzheimer’s disease (AD). Mary Hamby, PhD, and Lisa Ricciardi, CEO, Cognition Therapeutics Inc., Pittsburgh, PA, discuss the development of CT1812 and the evidence gathered to support its continued evaluation as a promising therapy for AD. First tested in a cortical neuronal culture system, CT1812 was shown to block toxic amyloid-beta (Aβ) oligomers from binding to neuronal synapses, restore trafficking deficits caused by oligomer accumulation, and protect synapses against damage. These findings were supported by studies in AD animal models that showed displacement of Aβ oligomers from synapses, reflected by increased Aβ in the cerebrospinal fluid. Improvements in cognition and memory deficits were also seen in rodents treated over several weeks. CT1812 has since completed three Phase I trials (SNAP, NCT03522129; SPARC, NCT03493282; and COG0102, NCT02907567) in which it was shown to be generally safe and tol
CT1812 is a small molecule sigma-2 receptor (S2R) modulator in development for Alzheimer’s disease (AD). Preclinical data from a cortical neuronal culture system and AD animal models have shown that CT1812 is able to displace amyloid-beta (Aβ) oligomers from synapses and prevent further receptor binding, thereby restoring damaged cellular processes such as autophagy and protein trafficking. Based on this mechanism of action, the team at Cognition Therapeutics wanted to see if S2R modulation could also block toxicity from alpha-synuclein (α-syn) oligomers. Preclinical testing demonstrated that CT1812 was able to block α-syn oligomers from binding to neuronal synapses, restore dysfunctional autophagy, and resolve protein trafficking deficits. CT1812 is therefore being investigated in the Phase II SHIMMER study (NCT05225415) in adults with dementia with Lewy bodies (DLB). This interview took place at the Alzheimer's Association International Conference (AAIC) 2022 in San Diego, CA.
Multi-center, randomized, double-blind, placebo-controlled, 6- month study in subjects with mild to moderate Dementia with Lewy Bodies.
This an open label study to assess the pharmacokinetics of CT1812 in normal healthy volunteers.
Open-label, single-dose study to assess the absorption, metabolism, excretion and mass balance of \[C14\] CT1812
This is a single-site, randomized, double-blind, placebo-controlled, 29-day, 2-period crossover Phase 2 study of 1 dose level of CT1812 (active) or placebo in adults with mild to moderate AD.
This is a multi-center, randomized, double-blind, placebo-controlled, parallel group 36 week multicenter Phase 2 study of two doses of CT1812 in adults with mild to moderate Alzheimer's Disease (AD).
This is a multi-center, Phase 1b, randomized, double-blind, placebo-controlled parallel-group trial in adults with mild to moderate AD.
Study to Evaluate the Safety and Tolerability of Oral CT1812 in Subjects with Mild to Moderate Alzheimer's Disease.
This is Phase 1, single-center, open-label, single-sequence drug-drug interaction study to determine the effect of CT1812 (560 mg) once daily on the pharmacokinetics of 4 probe drugs (tolbutamide, midazolam, dextromethorphan and omeprazole).
This is a multi-center, randomized, double-blind, placebo-controlled, parallel group study of two doses of CT1812 in adults with mild to moderate Alzheimer's Disease to evaluate the safety and tolerability of oral CT1812, administered for 28 days. This trial may include up to 8 qualified investigator sites in Australia.
This is a double-blind, placebo controlled, ascending dose, multi-cohort trial. The study will be conducted in two phases: a single ascending dose (SAD) phase "Part A", followed by a multiple ascending dose (MAD) phase "Part B". In Part A, subjects will receive one dose of study drug. In Part B, subjects within a cohort will receive the same dose daily for 14 days. In both parts, sequential cohorts will be exposed to increasing doses of CT1812 in order to identify the maximum tolerated dose (MTD).
The investigators will conduct a randomized placebo-controlled trial of a computerized intervention targeting cognition in 30 teens with autism spectrum disorder.
The purpose of this study is to determine the safety and efficacy of C105 in treating the cognitive deficits that can occur due to multiple sclerosis.
